چکیده
The dual mission of the Wisconsin Initiative for Science Literacy is to promote literacy in science, mathematics and technology among the general public and to attract future generations to careers in research, teaching and public service. At the March 5, 2010 UW-Madison Chemistry Department Colloquium, the director of the Wisconsin Initiative for Science Literacy (WISL) encouraged all Ph.D. chemistry candidates to include a chapter in their Ph.D. thesis communicating their research to non-specialists. The goal is to explain the candidate’s scholarly research and its significance to a wider audience that includes family members, friends, civic groups, newspaper reporters, state legislators, and members of the U.S. Congress.

19F NMR studies of the native and denatured states of green fluorescent protein.
Biosynthetic preparation and (19)F NMR experiments on uniformly 3-fluorotyrosine-labeled green fluorescent protein (GFP) are described. The (19)F NMR signals of all 10 fluorotyrosines are resolved in the protein spectrum with signals spread over 10 ppm. Each tyrosine in GFP was mutated in turn to phenylalanine. The spectra of the Tyr --> Phe mutants, in conjunction with relaxation data and resu...
متن کاملThe force-dependent mechanism of DnaK-mediated mechanical folding
It is well established that chaperones modulate the protein folding free-energy landscape. However, the molecular determinants underlying chaperone-mediated mechanical folding remain largely elusive, primarily because the force-extended unfolded conformation fundamentally differs from that characterized in biochemistry experiments. We use single-molecule force-clamp spectroscopy, combined with ...
متن کاملInteraction of epidermal growth factor with micelles monitored by photochemically induced dynamic nuclear polarization-1H NMR spectroscopy.
Photochemically induced dynamic nuclear polarization (CIDNP)-1H-NMR spectroscopy has been used to study the interaction of the protein hormone epidermal growth factor (EGF) with micelles of sodium dodecyl sulfate (SDS) and dodecylphosphorylcholine (DPC). Conventional 1H-NMR spectra show that most protein resonances remain unperturbed when micelles are added to solution, which argues that the ov...
متن کاملBinding of a small molecule at a protein-protein interface regulates the chaperone activity of hsp70-hsp40.
Heat shock protein 70 (Hsp70) is a highly conserved molecular chaperone that plays multiple roles in protein homeostasis. In these various tasks, the activity of Hsp70 is shaped by interactions with co-chaperones, such as Hsp40. The Hsp40 family of co-chaperones binds to Hsp70 through a conserved J-domain, and these factors stimulate ATPase and protein-folding activity. Using chemical screens, ...
متن کاملAtomic-Level Structure Characterization of an Ultrafast Folding Mini-Protein Denatured State
Atomic-level analyses of non-native protein ensembles constitute an important aspect of protein folding studies to reach a more complete understanding of how proteins attain their native form exhibiting biological activity. Previously, formation of hydrophobic clusters in the 6 M urea-denatured state of an ultrafast folding mini-protein known as TC5b from both photo-CIDNP NOE transfer studies a...
